Názov práce:The effect of apricot seeds on microscopic structure of rabbit ovaries
Abstrakt:Apricot (Prunus armeniaca L.) is a valuable fruit tree whose delicious fruits are widely used in the manufacture of various products (e.g. fruit juices, jams, etc.). Apricots are suitable for consumption both fresh and dried form but also their seeds may be used in various ways. In the dried form are consumed fresh apricot seeds, while bitter apricot seeds exercised its use as a raw material in the pharmaceutical and cosmetic industries. The bitter apricot seeds have high beta-carotene, magnesium, potassium, salicylic acid and folic acid content. Moreover, bitter apricot seeds contain a natural compound called amygdalin (also known as vitamin B17). This substance has been initiated for decades in alternative treatment of cancer therapy. The object of the study in presented thesis were apricot seeds (Prunus armeniaca L.) in order to determine the supplement and their potential effects on microscopic structure of the ovaries in model animals - rabbit. Ovaries as the central organ of the reproductive system of females among the most important organs in a living organism, that responds very sensitive on the various influences of the environment. In the in vivo experiment, we monitored potential changes in the microscopic structure of the ovaries of healthy female rabbits (n = 16) induced long-term (five months) administration of oral forms of apricot seeds, which we added to the feed at three selected doses of 60, 300 and 420 mg.kg-1 of body weight. We have seen the percentages of the various developmental stages of ovarian follicles after application of 60, 300 and 420 mg. kg.-1 of body weight doses of apricot seeds. In evaluating the percentage were observed significant reduction in the frequency of growing follicles (up to 2 layers of follicular cells) at a dose of 60 mg.kg-1 of body weight apricot seeds. Another significant difference which resulted in increase representation of ovarian follicles, we observed in the evaluation of the multiplicity of atretic follicles in the experimental group of females who have been fed with apricot seeds at a dose of 300 mg.kg.-1 of body weight. The growing presence of interstitium in ovarian microslices of experimental group contained the highest dose (400 mg.kg-1 of body weight) of apricot kernels has proved significant difference for the quantitative assessment of individual developmental stages of ovarian follicles. Further we focused on quantitative morphometric evaluation of individual follicular structures. Structural changes that have been shown to have significant were only registered in case of secondary follicle where they prove increased the diameter of oocytes in the experimental group, which have been fed with apricot seeds 300 mg.kg.-1 of body weight. Significant morphometric assessment was significant increase the diameter of the tertiary follicles in groups with doses of apricot kernel 60 and 300 mg.kg.-1 of body weight. All treatment groups were compared to the control group. In most reviews percentage of developmental stages of follicles and morphometric measurements of the diameters of the individual structures follicles we have not seen significant differences compared to the untreated control group. For these ratings can not explicitly declare the toxic effects resulting from the use of apricot seeds. Frequent inconclusive impact apricot seeds on microscopic structure of ovaries points out that the ovary as a body with autonomous protective barrier is not subject to changes , which could have classic histological examination alone document. Based on these results it can be concluded that 5 monthly addition of apricot seeds in feed experimentally groups of animals in three selected doses had not a clear negative effect on the microscopic structure of the ovaries of healthy female rabbits in vivo.
