Title of the thesis:Detection of genetic polymorphism prion protein gene (PRNP) by PCR method
Summary:Aim of the work was to identify the polymorphism PRNP gene and determine genotypic structure of populations Pinzgau cattle. Prions are infectious agents that are not in the genome of nucleic acids. Prion is able autoreplication and is composed almost entirely of normal cellular protein isoforms, which reproduces an unusual manner by converting to his own image similar existing protein found in all living cells. Prion is defined as a small protein infectious particle, which prevents inactivation by procedures that modify nucleic acids and cause fatal transmissible neurodegenerative diseases in humans and animals and are known as TSEs (transmissible spongiform encephalopathy). Biological material was obtained from 39 mothers of bulls of Slovak Pinzgau breed. Bovine genomic DNA was isolated by the method of proteolytic hydrolysis by proteinase K, phenol - chloroform and ethanol precipitation deprotenization and then was analyzed by PCR method to estimate PRNP genotypes. In the cattle population in the total number of 39 animals were found two genotypes: homozygous genotype 6/6, which had a predominance of genotype 5/6. Genotype responsible for the emergence of BSE 5/5 was not found.
