Molecular characteristics of selected species of cereals based on genetic diversity
Abstrakt:Diversity in plant genetic resources provides opportunity for plant breeders to develop new and improved cultivars with desirable characteristics. For genetic diversity analysis and accurate identification of genotypes is the most important to choose right molecular technique at the beginning, also determine the number of markers that is necessary to use for analysed set of genotypes. For determination of genetic diversity and identification of genotypes of wheat, rye, maize, oat and triticale three molecular techniques, RAPD, SCoT and STMS, were tested. Ability of identification and differentiation of genotypes, reproducibility of analysis and suitability of using of each technique for analysis of selected species of cereals was tested. RAPD technique was tested in the set of wheat, rye and maize genotypes and revealed high polymorphism. Genotypes of maize were distinguished using 13 RAPD markers and genotypes of wheat using 6 RAPD markers. In the analysis of rye, rye genotypes using only 10 RAPD primers grouped partially with regard to country of origin in the dendrogram. Using SCoT technique sets of maize and oat genotypes were tested. SCoT technique revealed sufficiently polymorphic for identification and differentiation of sets of maize and oat genotypes. STMS technique was used for molecular analysis of a set of wheat, rye and triticale genotypes. Exploitation of wheat SSR markers for genetic diversity analysis of wheat, also utilization of wheat and rye SSR markers in the analysis of triticale and rye SSR markers for analysis of rye was confirmed. Higher level of polymorphism was detected by using of wheat SSR markers compared to rye SSR markers in triticale. Only 6 SSR markers were able to differentiate all analysed rye genotypes. Based on the analysis results that for discrimination of all genotypes in the chosen set of genotypes it is necessary to choose mainly high polymorphic markers and low number of markers is sufficient. Used molecular techniques (RAPD, SCoT, STMS) revealed sufficient polymorphism for differentiation and identification of analysed genotypes of cereals. All three techniques were reproducible. Marker index which defines general marker ability to detect polymorphism, reached the highest values in the analysis using RAPD technique for the detection of genetic diversity of rye and maize. For the estimation of sufficient number of marker loci is used diversity detecting index (DDI). The highest values of DDI reached SCoT technique in the analysis of a set of oat and maize genotypes.
